Sequencing Assay Overviews
Choose a Cloudbreak™, Cloudbreak Freestyle™, Cloudbreak UltraQ™, or Trinity™ kit before you plan a sequencing run.
A sequencing run uses one of these kits on an AVITI System or an AVITI24™ System. The kit decides which libraries you can load and whether the run is standard sequencing or onboard enrichment. Choose a kit in the table. Then follow the end-to-end map to plan the run, prepare a manifest, sequence, and demultiplex to FASTQ.

Cloudbreak
Cloudbreak offers the same read lengths and output options as Cloudbreak Freestyle. Some libraries require circularization before loading. In ElemBio Cloud, plan the run as Sequencing with Cloudbreak and select Adept or Elevate.
Adept libraries use the Adept workflow. On original Cloudbreak chemistry, replace the cartridge primers with the Adept primer set. Elevate libraries use Elevate indexes and adapters. The primers in the cartridge are compatible with those libraries.

Cloudbreak Freestyle
Cloudbreak Freestyle loads linear libraries directly, without library conversion, including third-party libraries. In ElemBio Cloud, plan the run as Sequencing with Cloudbreak and select Third Party.
The Adept primer set is not compatible with Cloudbreak Freestyle kits.

Cloudbreak UltraQ
Cloudbreak UltraQ provides high-quality Q40 and Q50 data for sensitive assays. The Adept primer set is not compatible with Cloudbreak UltraQ kits. Plan the run as Sequencing with Cloudbreak.

Trinity
Trinity is a separate planned-run type for onboard enrichment on an AVITI System or an AVITI24 System. Select Elevate or Third Party when you plan a Trinity run. That page lists the panels and cycle limits for the kit you select.
Prepare the manifest from the templates on Prepare a sequencing manifest. Use the Elevate Trinity template for Elevate libraries, or an Adept or third-party template for a third-party library.
